ABSTRACT
Cigarette smoke contains free radicals and cause free radicals formation in human body. The excess of free radicals in the body will be increased lipid peroxidation and destroy the cell wall. Beside that, human body has a protective mechanism through free radicals “scavenger”, such as superoxide dismutase (SOD). The purpose of this study is to determine the correlation between the axcess of free radicals activity of “kretek” cigarette smoke and malondyaldehide (MDA) as a lipid peroxidation indicator with SOD as a protector system in Wistar Rat Liver. This research was done in Biomedical Laboratory, Faculty of Medicine, Brawijaya University, Malang. Experimental design used five group of Wistar rat and each group was 8 wistar rat. The first group used as a control from the second group to fifth group were exposed with two “kretek” cigarette smoke daily for 2, 4, 8 and 10 weeks, respectively. Malondylaldehid (MDA) level of rat liver homogenat were measured by used Thiobarbituric Acid (TBA) methods from Uchiyama and Mihara. SOD was determined by followed Wong?s protocol. This research results were showed that “kretek” cigarette smoke can increase MDA level from 0,17 ppm (control group) to 0,3 ppm (2 week group), 0,5 ppm (4 week group), 0,76 ppm (8 week group) and 2,34 ppm (10 week group). (Anava significant P < 0,005) and also decrease SOD from 47,66 U/mg (control group) to 6,61 U/mg (2 week group), 3,89 U/mg (4 week group), 1,52 U/mg (8 week group), and 0,15 ppm (10 week group) in rat liver (Anava significant P < 0,005). We conclude that “kretek” cigarette smoke can be increased free radicals and decreasd superoxide dismutase as enzymatic protector system in Wistar rat liver.
